RESUMO

Accurate and specific imaging of low-abundance microRNA (miRNA) in living cells is extremely important for disease diagnosis and monitoring of disease progression. DNA nanomotors have shown great potential for imaging molecules of interest in living cells. However, inappropriate driving forces and complex design and operation procedures have hindered their further application. Here, we proposed an endogenous enzyme-powered DNA nanomotor (EEPDN), which employs an endogenous APE1 enzyme as fuel to execute repetitive cycles of motion for miRNA imaging in living cells. The whole motor system is constructed based on gold nanoparticles without other auxiliary additives. Due to the high efficiency of APE1, this EEPDN system has achieved highly sensitive miRNA imaging in living cells within 1.5 h. This strategy was also successfully used to differentiate the expression of specific miRNA between tumor cells and normal cells, demonstrating a high tumor cell selectivity. This strategy can promote the development of novel nanomotors and is expected to be a perfect intracellular molecular imaging tool for biological and medical applications.

Ovarian cancer is the second most common gynecological malignancy, and one of the most deadly. The bottleneck restricting the treatment of ovarian cancer is its multi-drug resistance to chemotherapy. Cajanol is an isoflavone from pigeon pea (Cajanus cajan) that has been reported to have anti-tumor activity. In this work, we evaluate the effect of cajanol in reversing paclitaxel resistance of the A2780/Taxol ovarian cancer cell line in vitro and in vivo, and we discuss its mechanism of action. We found that 8 µM cajanol significantly restored the sensitivity of A2780/Taxol cells to paclitaxel, and in vivo experiments demonstrated that the combination of 0.5 mM/kg paclitaxel and 2 mM/kg cajanol significantly inhibited the growth of A2780/Taxol metastatic tumors in mice. Flow cytometry, fluorescence quantitative PCR, western blotting and immunohistochemical staining methods were used to study the mechanism of reversing paclitaxel resistance with cajanol. First, we determined that cajanol inhibits paclitaxel efflux in A2780/Taxol cells by down-regulating permeability glycoprotein (P-gp) expression, and further found that cajanol can inhibit P-gp transcription and translation through the PI3K/Akt/NF-κB pathway. The results of this work are expected to provide a new candidate compound for the development of paclitaxel sensitizers.

The removal of cadmium(ii) pollution in surface water by a polymer enhanced ultrafiltration (PEUF) process was investigated. Three water soluble polymers, chitosan (CTS), polyvinyl alcohol (PVA) and polyacrylate sodium (PAAS), were selected for this study. Of the three polymers, PAAS had strong interactions with Cd2+, and the PEUF achieved a high removal of Cd2+; therefore, PAAS was used as a complexing agent for the simulated cadmium pollution experiment. Experiments were performed as a function of the aqueous pH, polymer/Cd2+ ratio (P/M), ionic strength and humic acid. Under optimum experimental conditions, the Cd2+ removal rate reached 100%. pH was the main factor affecting removal of Cd2+, which decreased to 60% at a pH of 4. The Cd2+ removal was found to decrease as NaCl and HA were added. The analysis showed that the mechanism of NaCl could be a compressed electric double layer, while the mechanism of HA and H+ was competitive complexation. Finally, UF membrane fouling, the dissociation of PAAS-Cd and the regeneration of PAAS were investigated. The results showed that the higher P/M was, the lower the pH, the higher the HA concentration, and the more serious the UF membrane fouling were. The dissociation rate of PAAS-Cd reached 99.8% at a pH of 2.5. When the P/M was 5, the removal rate of Cd reached 99.6% with the addition of 20% new PAAS in the regenerate. This result showed that the PEUF process could be a promising method for removing Cd pollution in surface water.
